Many unnaturally modified nucleobases, such methyl, halogen, and aryl modifications of purines at the C8 position and pyrimidines during the C5 position, are extensively examined with regards to their biological functions. DNA containing these customized nucleobases could form non-canonical helical structures such as for instance Z-DNA, G-quadruplex, i-motif, and triplex. This review summarizes the formation of chemically changed nucleotides (i) methylation, bromination, and arylation of purine at the C8 position and (ii) methylation, bromination, and arylation of pyrimidine during the C5 place.
