Deterministic and stochastic processes are believed to be interwoven in the formation of microbial communities, although the determining elements of their respective contributions remain elusive. Using biofilm carriers with precisely controlled maximum biofilm thickness, we examined how biofilm thickness impacts community assembly within nitrifying moving bed biofilm reactors. A steady-state biofilm assembly was scrutinized for stochastic and deterministic contributions using neutral community modelling and a null-model analysis of community diversity. Our investigation indicates that biofilm formation leads to habitat filtration, favoring phylogenetically related community members. This process contributes to a substantial increase in the number of Nitrospira spp. observed within the biofilm communities. Biofilms thicker than 200 micrometers displayed a greater prevalence of stochastic assembly processes, in contrast to the 50-micrometer-thick biofilms, where surface hydrodynamic and shear forces facilitated stronger selection. Anti-idiotypic immunoregulation More substantial biofilms exhibited a greater degree of phylogenetic beta-diversity; this could stem from varying selective pressures triggered by environmental differences across replicate communities, or from a mix of genetic drift and reduced migration, leading to unpredictable events during community assembly. Our findings suggest that biofilm assembly procedures are contingent upon the thickness of the biofilm, which enhances our comprehension of biofilm ecology and may potentially open avenues for managing microbial communities within biofilm systems.
